    Transcript expression of vesicular glutamate transporters in lumbar dorsal root ganglia and the spinal cord of mice – Effects of peripheral axotomy or hindpaw inflammation

    Using specific riboprobes, we characterized the expression of vesicular glutamate transporter (VGLUT)1–VGLUT3 transcripts in lumbar 4–5 (L4–5) dorsal root ganglions (DRGs) and the thoracolumbar to lumbosacral spinal cord in male BALB/c mice after a 1- or 3-day hindpaw inflammation, or a 7-day sciatic nerve axotomy. Sham animals were also included. In sham and contralateral L4–5 DRGs of injured mice, VGLUT1-, VGLUT2- and VGLUT3 mRNAs were expressed in ∌45%, ∌69% or ∌17% of neuron profiles (NPs), respectively. VGLUT1 was expressed in large and medium-sized NPs, VGLUT2 in NPs of all sizes, and VGLUT3 in small and medium-sized NPs. In the spinal cord, VGLUT1 was restricted to a number of NPs at thoracolumbar and lumbar segments, in what appears to be the dorsal nucleus of Clarke, and in mid laminae III–IV. In contrast, VGLUT2 was present in numerous NPs at all analyzed spinal segments, except the lateral aspects of the ventral horns, especially at the lumbar enlargement, where it was virtually absent. VGLUT3 was detected in a discrete number of NPs in laminae III–IV of the dorsal horn. Axotomy resulted in a moderate decrease in the number of DRG NPs expressing VGLUT3, whereas VGLUT1 and VGLUT2 were unaffected. Likewise, the percentage of NPs expressing VGLUT transcripts remained unaltered after hindpaw inflammation, both in DRGs and the spinal cord. Altogether, these results confirm previous descriptions on VGLUTs expression in adult mice DRGs, with the exception of VGLUT1, whose protein expression was detected in a lower percentage of mouse DRG NPs. A detailed account on the location of neurons expressing VGLUTs transcripts in the adult mouse spinal cord is also presented. Finally, the lack of change in the number of neurons expressing VGLUT1 and VGLUT2 transcripts after axotomy, as compared to data on protein expression, suggests translational rather than transcriptional regulation of VGLUTs after injury.Fil: Malet, Mariana.     Topological transition in measurement-induced geometric phases

    The state of a quantum system, adiabatically driven in a cycle, may acquire a measurable phase depending only on the closed trajectory in parameter space. Such geometric phases are ubiquitous and also underline the physics of robust topological phenomena such as the quantum Hall effect. Equivalently, a geometric phase may be induced through a cyclic sequence of quantum measurements. We show that the application of a sequence of weak measurements renders the closed trajectories, hence the geometric phase, stochastic. We study the concomitant probability distribution and show that, when varying the measurement strength, the mapping between the measurement sequence and the geometric phase undergoes a topological transition. Our finding may impact measurement-induced control and manipulation of quantum states-a promising approach to quantum information processing. It also has repercussions on understanding the foundations of quantum measurement

    True aneurysm of the peripheral pulmonary artery due to necrotizing giant cell arteritis

    Pulmonary artery aneurysm in adults is a rare diagnosis. Most cases described in the literature are either associated with congenital heart disease or pulmonal arterial hypertension, respectively, or are not true aneurysms but rather pseudoaneurysms, which are usually iatrogenic. We present the case of a 68-year old female patient with the incidental finding of a true aneurysm of the right peripheral pulmonary artery with a maximum diameter of 4 cm. With increasing aneurysm diameter over time, the decision for a surgical resection was made. Complete resection of the aneurysm including lower lobe resection was performed. Histopathological examination showed necrotizing giant cell arteritis as the underlying cause. The postoperative course was uneventful and no signs of further disease activity were detected. To our knowledge, this is the first reported case of a pulmonary artery aneurysm caused by giant cell arteritis, whereas it should be noted that the distinction between Takayasu arteritis and giant cell arteritis is not clearly defined. Considering the high mortality associated with aneurysm rupture, surveillance is advocated for small aneurysms, whereas for larger aneurysms and those showing signs of progression in size despite medical therapy or even dissection, surgical intervention should be considere

    Optically thin cooling gas at most temperatures above 30K will make condensations by pressure pushing material into cool dense regions. This works without gravity. Cooling condensations will flatten and become planar/similarity solutions. Most star formation may start from cooling condensations - with gravity only important in the later stages. The idea that some of the dark matter could be pristine white dwarfs that condensed slowly on to planetary sized seeds without firing nuclear reactions is found lacking. However, recent observations indicate fifty times more halo white dwarfs than have been previously acknowledged; enough to make the halo fraction observed as MACHOS.     Learning Quantum Systems

    Quantum technologies hold the promise to revolutionise our society with ground-breaking applications in secure communication, high-performance computing and ultra-precise sensing. One of the main features in scaling up quantum technologies is that the complexity of quantum systems scales exponentially with their size. This poses severe challenges in the efficient calibration, benchmarking and validation of quantum states and their dynamical control. While the complete simulation of large-scale quantum systems may only be possible with a quantum computer, classical characterisation and optimisation methods (supported by cutting edge numerical techniques) can still play an important role. Here, we review classical approaches to learning quantum systems, their correlation properties, their dynamics and their interaction with the environment. We discuss theoretical proposals and successful implementations in different physical platforms such as spin qubits, trapped ions, photonic and atomic systems, and superconducting circuits. This review provides a brief background for key concepts recurring across many of these approaches, such as the Bayesian formalism or Neural Networks, and outlines open questions

    New Class of Precision Antimicrobials Redefines Role of Clostridium difficile S-layer in Virulence and Viability

    There is a medical need for antibacterial agents that do not damage the resident gut microbiota or promote the spread of antibiotic resistance. We recently described a prototypic precision bactericidal agent, Av-CD291.2, which selectively kills specific Clostridium difficile strains and prevents them from colonizing mice. We have since selected two Av-CD291.2–resistant mutants that have a surface (S)-layer–null phenotype due to distinct point mutations in the slpA gene. Using newly identified bacteriophage receptor binding proteins for targeting, we constructed a panel of Avidocin-CDs that kills diverse C. difficile isolates in an S-layer sequence-dependent manner. In addition to bacteriophage receptor recognition, characterization of the mutants also uncovered important roles for S-layer protein A (SlpA) in sporulation, resistance to innate immunity effectors, and toxin production. Surprisingly, S-layer–null mutants were found to persist in the hamster gut despite a complete attenuation of virulence. These findings suggest antimicrobials targeting virulence factors dispensable for fitness in the host force pathogens to trade virulence for viability and would have clear clinical advantages should resistance emerge. Given their exquisite specificity for the pathogen, Avidocin-CDs have substantial therapeutic potential for the treatment and prevention of C. difficile infections
